Stability Testing for Biotinylated Recombinant Human CD40L: Biotinylated recombinant human CD40L lyophilized was kept at -20°C (Control), and another sampler was reconstituted in PBS, pH7.2 and kept at 4°C for two weeks (2w 4°C). The samples were tested by their property to bind to immobilized recombinant human CD40-Fc chimera (Cat. No. 777204) in a dose-dependent manner. The ED50 for this effect is 0.6 – 4.0 ng/mL.

CD40L is a type II membrane protein that shares 78% amino acid identity with its mouse counterpart. Human CD40L is a polypeptide of 261 amino acids, consisting of 22, 24, and 215 amino acids for the cytoplasmic, transmembrane, and extracellular domains respectively. Soluble forms of CD40L (sCD40L) have been identified, and an 18 kD fragment can form homotrimers and bind to CD40 to induce biological signals. The 18 kD protein has full activity in B cell proliferation and differentiation assays. Activated platelets express CD40L on their surface and constitute the major source of sCD40L (>95% in blood). Platelet-derived CD40L regulates neutrophil recruitment and lung damage in sepsis, and it has been suggested that MMP9 regulates CD40L shedding in abdominal sepsis. Platelet-associated CD40L has been linked to progression of atherosclerotic disease, and elevated plasma levels of sCD40L in patients seems to be a reliable predictor of cardiovascular events, such as atherosclerotic plaque rupture and acute coronary syndromes. Also, platelet-associated CD40L expression is increased in idiopathic thrombocytopenic purpura patients, and it has been speculated that platelet-associated-CD40L drives the activation of autoreactive B lymphocytes in this disease. In addition, high plasma levels of sCD40L have been connected to HIV-Associated Neurocognitive Disorders (HAND).

- Human CD40L, amino acids Gly116-Leu261 (Accession# P29965) was expressed in 293E cells. The N-terminal contains an Avi-8His tag. Human CD40L-Avi tag was site-specifically biotinylated by enzyme BirA.

- The 192 amino acid recombinant protein has a predicted molecular mass of approximately 20.6 kD. The protein migrates at approximately 27 - 30 kD in DTT-reducing and non-reducing conditions by SDS-PAGE. The predicted N-terminal amino acid is Gly.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C or colder until the expiration date. Reconstitute lyophilized protein in sterile PBS, pH7.2. Before reconstitution, make sure sterile PBS and product are at room temperature. Quickly spin the vial or gently tap down on the vial to make sure the lyophilized product is at the bottom of the vial before opening. Use aseptic techniques to add the required volume of reconstitution buffer (sterile PBS) to the vial, to obtain the recommended stock concentration 250 µg/mL. Close the vial and leave at an ambient temperature for 15-20 minutes. Then gently invert the vial several times or until all the lyophilized product dissolves. Leave the vial at room temperature for another 15 minutes. If small particulates are still observed after 15 minutes, incubate at room temperature for an additional 30 minutes, and leave the vial at 2°C - 8°C overnight. Next day, invert the vial several times or gently pipette the solution up and down before use. If needed, transfer the reconstituted stock solution to a sterile container for additional dilution to no less than 100 µg/mL. Small working aliquots in polypropylene tubes can be made after reconstitution and store the vials at -20°C or lower. Avoid freeze/thaw cycles. Carrier protein such as 0.2 - 1% endotoxin-free BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to two weeks or stored at -20°C or colder for up to 3 months.

- CD40L induces proliferation, differentiation, and immunoglobulin class switching in B cells; also prevents apoptosis in B cells. CD40L is induced in T cells after in vitro activation with PHA, PMA plus ionomycin, anti-CD3, anti-CD2, and CD28 engagement in anti-CD3 activation. Also, CD40L is induced by thrombin in platelets.
