IHC staining with Purified anti-ZO-2 antibody on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) and with (panel B) Purified anti-ZO-2 antibody (clone W23248A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 50 μm

    A-431 cells (top panels, positive control) or A-549 cells (bottom panels, negative control) were fixed and permeabilized with PFA and triton X-100, and then intracellularly stained with Purified anti-ZO-2 (clone W23248A) (right panels) or Purified Rat IgG2a,  Isotype Control (Cat No. 400502)(left panels) followed by Alexa Fluor® 647 Goat anti-rat IgG (Cat.     Whole cell lysates (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-ZO-2 antibody (clone W23248A). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG antibody (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control. Description

The ZO-2 protein is a key component of tight junctions. It plays a role in maintaining cell polarity and barrier function. ZO-2 interacts with other tight junction proteins like claudins and occludins and participates in signaling pathways that influence gene expression and cell proliferation. Its expression is regulated by cellular stress, cytokines, and developmental cues. Mutations in TJP2 have been associated with several diseases, including progressive familial intrahepatic cholestasis type 4 (PFIC4), familial hypercholanemia, and autosomal dominant non-syndromic hearing loss (DFNA51).

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).

For use of this antibody in Immunocytochemistry (ICC), we recommendf ixation and permeabilization using PFA-Triton X-100 or PFA-ice-cold methanol. We do not recommend ice-cold methanol only.
