Whole cell extracts (15 µg total protein per lane) from indicated cells and cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Direct-Blot™ HRP anti-E. Coli RNA Sigma 70 Recombinant (clone 2G10.RecM1) overnight at 4°C. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426315) was used as a detection agent. Most prokaryotes such as E. coli express a single core RNA polymerase to transcribe RNA from DNA. RNA Sigma 70, also known as rpoD, is the major housekeeping member of the sigma factor subunit family which are required for core RNA polymerase to bind to promoters and initiate transcription. RNA sigma 70 is also required for the unwinding of DNA strands to allow access to RNA polymerase.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 1:1000 – 1:4000. It is recommended that the reagent be titrated for optimal performance for each application.

This recombinant antibody is derived from the original clone 2G10. We sequenced the complementarity-determining regions (CDRs) from the 2G10 hybridoma and grafted them into a mouse IgG1, κ Fc framework. Clone 2G10 specifically recognizes the major sigma factor across multiple bacterial species but shows no reactivity against Borrelia burgdorferi. Clone 2G10 is known to target epitope 470-486 in the major sigma factor of E. coli.
