    HeLa cells were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then either stained with Purified Rat IgG2b, κ Isotype Control (Cat. No. 400602) (Panel A) or Purified anti-SMC3 (clone W23212B) (Panel B), followed by incubation with Alexa Fluor® 647 Goat anti-rat (Cat. No. 400602) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm

Description

SMC3 (Structural Maintenance of Chromosomes 3) is a core component of the cohesin complex, a group of proteins essential for chromosome cohesion, segregation, DNA repair, and transcriptional regulation in eukaryotic cells. SMC3 belongs to the Structural Maintenance of Chromosomes (SMC) family of ATPases, which includes other members such as SMC1, SMC2, SMC4, SMC5, and SMC6. Together with SMC1A, RAD21 (SCC1), and STAG proteins (SA1/SA2), SMC3 forms the ring-shaped cohesin complex that topologically encircles sister chromatids, holding them together after DNA replication until their separation during mitosis or meiosis. SMC3 and the broader cohesin complex are involved in regulating gene expression through chromatin architecture remodeling. Cohesin interacts with CTCF (CCCTC-binding factor), a key architectural protein, to form chromatin loops and topologically associating domains (TADs). These structures bring enhancers and promoters into close proximity, modulating transcriptional programs necessary for development and differentiation.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:
-Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
