Indicated cell lysates (15 µg total protein) were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-MCL-1 antibody (clone W23229E) for overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
SR cells (high-expressing cell line, panel B) and Jurkat cells (low-expressing cell line, panel A) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-Mcl-1 (clone W23229E) and Purified mouse anti-PDHA1 (clone 3H2-F8-B5, Cat. No. 610951), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) and Alexa Fluor® 488 Goat anti-mouse IgG (Cat. No. 405319) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm

Mcl-1 is a member of the pro-survival BCL-2 family of apoptotic regulatory proteins. Mcl-1 plays a crucial role in the survival and development of various cell types by binding to proapoptotic proteins and inhibiting the mitochondrial apoptotic pathway. Unlike Bcl-2 and Bcl-xL, Mcl-1 lacks a BH4 domain and contains an N-terminal PEST region with specific sequences for ubiquitination, phosphorylation, and caspase cleavage sites, which are important in regulating the half-life. Phosphorylation of at T92 and T163 by ERK inhibits Mcl-1 turnover, whereas phosphorylation by GSK3β at S159 facilitates proteosomal degradation. Increased expression of Mcl-1 is correlated with tumor survival and is associated with resistance of chemotherapeutics.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
