Whole cell lysates (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-LMO2 (clone W23203A). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG antibody (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
Jurkat cells (negative control, top panels) or TF-1 cells (positive control, bottom panels) were fixed and permeabilized with PFA and ice-cold methanol, and then intracellularly stained with Purified anti-LMO2 (clone W23203A) (right panels) or Purified Rat IgG2a Isotype Control (left panels) (Cat. No. 400502) followed by Alexa Fluor® 647 Goat anti-rat IgG (Cat No. 405416). Cells were counterstained with DAPI (Cat. No. 422801) (blue). Images were captured using a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm -
TF-1 cells (positive cell line, filled histogram) and Jurkat cells (negative cell line, open histogram) were fixed and permeabilized using True-Phos™ Perm Buffer (Cat. No. 425401) and intracellularly stained with Purified anti-LMO2 (clone W23203A), or Purified Rat IgG2a, κ Isotype Control (dashed histogram, representative for both cell lines) (Cat. LMO2 (LIM Domain Only 2) is a cysteine-rich protein containing two LIM domains, playing a pivotal role in hematopoietic development. It functions primarily as a transcriptional regulator, forming multiprotein complexes with other transcription factors such as TAL1/SCL and LDB1 to control gene expression critical for erythropoiesis and the maintenance of hematopoietic stem and progenitor cells. LMO2 is tightly regulated at the transcriptional level, with its expression modulated by various enhancers and transcription factors, including AML1a and SP1. Dysregulation of LMO2, particularly its ectopic expression in T cells, is strongly associated with T-cell acute lymphoblastic leukemia (T-ALL), where chromosomal translocations involving the LMO2 locus led to aberrant activation. Additionally, LMO2 has been implicated in other hematologic malignancies and may act as a proto-oncogene, highlighting its dual role in normal development and disease pathogenesis.

ICC, ICFC - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.03 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401), or True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401). We do not recommend Cyto-Fast™ Fix-Perm Buffer set (Cat. No. 426803).
For use of this antibody in immunocytochemistry (ICC), we recommend fixation and permeabilization using either:
-PFA then ice cold methanol
-PFA then Triton X-100
