C57BL/6 mouse splenocytes were stimulated with plate-bound Ultra-LEAF™ purified anti-mouse TCR β chain (Cat. No. 109254) for 2 days, and surface stained with anti-mouse CD8a (clone 53-6.7) PE. Cells were then fixed and permeabilized with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) and intracellularly stained with anti-mouse Granzyme B (clone S23006A) Alexa Fluor® 647 (left) or rat IgG1, λ Alexa Fluor® 647 isotype control (right).

Granzyme B, also known as granzyme-2, serine protease B, CCP1, Asp-ase, and CTLA-1, is a 32 kD serine protease. Granzyme B is abundantly stored in the granules of cytotoxic T lymphocytes and NK cells. Low level of expression has been reported in granulocytes, B cells, and activated dendritic cells. Granzyme B is crucial for rapid induction of cell death and apoptosis through interaction with mannose-6-phosphate receptor.

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.
