Direct ELISA showing biotin anti-human YKL-40 (CHI3L1) (clone A19120F) bound to plate-immobilized recombinant human YKL-40 (CHI3L1). The wells were incubated with serially diluted primary biotin antibody at room temperature for 2 hours in 1% BSA-PBS, followed by incubation with HRP Avidin (Cat No. 405103). TMB High Sensitivity Substrate Solution (Cat No. 421501) was used for detection. Absorbance was measured at 450 nm.

Description

YKL-40, also known as CHI3L1 (Chitinase-3-like protein 1) or cartilage glycoprotein-39 (GP-39) is secreted by chondrocytes, synoviocytes, endothelial cells, smooth muscle cells and immune cells. YKL-40 plays a major role in different physiological processes including cell proliferation, tissue remodeling, angiogenesis, and apoptosis. Elevated serum levels of YKL-40 are linked to the pathogenesis and prognosis of several inflammatory reactions and autoimmune diseases, such as cancer, rheumatoid arthritis, liver cirrhosis, diabetes, and inflammatory bowel disease. Higher YKL-40 levels in cerebral spinal fluid are a known biomarker of brain inflammation and have a potential prognostic utility as a disease-specific marker in neurodegenerative disorders.

Additional Product Notes

This product is intended for sandwich-based immunoassay development on various assay platforms requiring antibody pairs. This antibody (clone A19120F) could function as a detection antibody when paired with purified anti-human YKL-40 (CHI3L1) antibody (Cat No. TBD) (clone W19120B) as the capture antibody and using Recombinant Human Chitnase 3-like 1 (carrier-free) (Cat No. 56802 and 568706) protein as the assay standard. For assay development flexibility purposes, this product is provided as a biotin format.
