IHC staining with Purified anti-FABP2 (clone W24079A) on formalin-fixed paraffin-embedded human colon tissue. Following antigen retrieval using 1X Citrate Buffer (Cat. No. 420902), the tissue was incubated with Purified anti-FABP2 (clone W24079A) (panel A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged (panel B). Scale bar: 50 µm -
Whole cell extracts (15 µg total protein per lane) from indicated tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-FABP2 (clone W24079A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Fatty acid-binding protein 2 (FABP2), also known as intestinal fatty acid-binding protein (I-FABP), is a small intracellular lipid-binding protein primarily expressed in the enterocytes of the small intestine. FABP2 belongs to the family of fatty acid-binding proteins that play crucial roles in lipid metabolism by binding long-chain fatty acids and other hydrophobic ligands. FABP2 is involved in the uptake, transport, and intracellular trafficking of dietary fatty acids from the intestinal lumen to metabolic or storage pathways, acting as a lipid chaperone. It contributes significantly to the regulation of lipid absorption and energy homeostasis. A polymorphism in FABP2, Ala54Thr (rs1799883), results in an amino acid substitution at position 54 and has been extensively studied for its metabolic implications. This variant is linked to several metabolic disorders, including insulin resistance, type 2 diabetes mellitus (T2DM), dyslipidemia, and obesity. In cardiovascular disease, FABP2 variants and expression levels are associated with altered lipid profiles and atherogenic risk, likely due to disrupted lipid transport from the gut to the circulation.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
