Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-EHF (clone W22125A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:5000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

W22125A_PURE_EHF_ICC_62425
    LNCaP cells with (EHF KD) and without (mock KD) transient transfection of EHF shRNA were fixed with fixation buffer (Cat. No. 420801) for 10 minutes, permeabilized with ice-cold methanol, and then blocked with 5% FBS for 1 hour at room temperature. Cells were then intracellularly stained with isotype control (Cat. No. 400602) (panels A and D) or stained with Purified anti-EHF (clone W22125A) (panels B and E), followed by incubation with Alexa Fluor 594 Goat anti-rat IgG (red, Cat. No. 405422) for 1 hour at room temperature. Then, cells were co-stained with Alexa Fluor® 488 anti-GFP Antibody (green, Cat. No. 338008) (merged in panels C and F) for 1 hour at room temperature to detect the knockdown reporter in transfected cells. Nuclei were counterstained with DAPI (Cat. No. 422801) (merged in panels C and F). The image was captured with a 40X objective. Scale bar: 25 µm

Description

ETS homologous factor (EHF), also known as epithelium-specific ETS transcription factor 3 (ESE-3), is a member of the ETS family of transcription factors expressed in various epithelial cell types, which participates in the oncogenesis of many malignant tumors through regulating the cell proliferation and differentiation. The genome-wide association study (GWAS) identified that genetic markers of lung disease severity in cystic fibrosis (CF) led to renewed interest in the potential importance of EHF in lung disease. Studies have revealed that EHF transcript variants such as EHF-LF and EHF-SF may have different impacts on epithelial cell differentiation and can promote or inhibit the epithelial mesenchymal transition (EMT) phenomenon. EHF also has been found in kidney, lung and somatotroph tumors and may serve as a prognostic biomarker. According to recent studies, EHF has been shown to inhibit pancreatic ductal adenocarcinoma (PDAC) metastasis through up-regulation of E-cadherin.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For immunocytochemistry (ICC), we recommend 4% PFA fixation followed by permeabilization with Triton X-100 or ice-cold methanol.

This antibody (clone W22125A) is not suitable for IHC.

This antibody (clone W22125A) does not cross-react with mouse in western blot (WB).
