Description

Carbonic anhydrase 2 (CA2) is an enzyme that catalyzes the reversible hydration of carbon dioxide, playing a role in maintaining acid-base balance and facilitating CO2 transport in the blood. It is regulated by various factors, including the availability of zinc ions, which are essential for its catalytic activity. CA2 is involved in several physiological processes, such as respiration and pH regulation in tissues. Defects in CA2 can lead to diseases like osteopetrosis, a condition characterized by overly dense bones, and renal tubular acidosis, which affects the kidney ability to maintain acid-base balance. Additionally, CA2 deficiency can result in brain calcifications and developmental delays.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401). We do not recommend True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401) or Cyto-Fast™ Fix-Perm Buffer set (Cat. No. 426803).

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).

Antigen Details

Structure
Carbonic anhydrase 2 (CA2) consists of 260 amino acids and has a molecular weight of approximately 29.2 kDa.
Distribution

Carbonic anhydrase 2 (CA2) is primarily located in the cytoplasm

Function
• Catalyzes CO2 Hydration: Converts carbon dioxide (CO2) to bicarbonate (HCO3-) and protons (H+), facilitating CO2 transport and maintaining acid-base balance.
• pH Regulation: Helps regulate pH in various tissues, including red blood cells, kidneys, and the gastrointestinal tract.
• Respiratory Function: Plays a crucial role in the rapid exchange of CO2 and oxygen in the lungs
• Bone Resorption: Involved in bone resorption by osteoclasts, essential for bone remodeling.
Interaction
• Band 3 (AE1): The erythrocyte anion exchanger involved in chloride/bicarbonate exchange.
• Sodium-Hydrogen Antiporter 1 (NHE1): A protein that helps regulate intracellular pH by exchanging sodium ions for hydrogen ions across the cell membrane.

Regulation
• Zinc Ion Availability: Requires zinc ions for its catalytic activity; zinc binding is crucial for enzyme function.
• Gene Expression: Regulated at the transcriptional level by various factors, including hormones and cellular pH.
• Inhibitors: Can be inhibited by specific drugs, such as acetazolamide, which are used to treat conditions like glaucoma and epilepsy
