Stability testing for Biotinylated human FAP: Biotinylated human FAP was aliquoted in 16 mM Tris, 240 mM NaCl, 20% Glycerol, pH 7.5 at 0.2 mg/mL and one aliquot was kept at 4°C (Control) for 7 days and another aliquot was frozen and thawed four times (4x Freeze/Thaw) over a period of one week. After this procedure, samples were tested for their ability to cleave a fluorogenic substrate Z-Gly-Pro-AMC with a specific activity value > 1800 pmol/µg/min.

Fibroblast activation protein (FAP) is a cell-surface transmembrane-anchored dimeric proteas. FAP expression is very low in adult organs but is upregulated by activated fibroblasts at the sites of tissue remodeling, including fibrosis, atherosclerosis, arthritis and tumors. FAP is broadly expressed on cancer associated fibroblasts (CAFs), which are a major component of the stromal microenvironment of carcinomas. FAP has been identified as a promising immunotherapy target antigen for epithelial carcinomas such as prostate, lung, pancreatic cancer and mesothelioma. It has been shown that by removing the fibrosis barrier, ‘FAP-Chimeric Antigen Receptor-Macrophages’ enhanced the penetration of gemcitabine and immune cells, improved PDAC (pancreatic ductal adenocarcinoma) sensitivity to chemotherapy and immunotherapy and significantly prolonged survival. FAP is not commonly expressed by blood vessels within healthy tissues or by normal endothelial cells (ECs) but can be strongly and consistently induced on ECs within the glioblastoma microenvironment. FAP directly cleaves CXCL12 to inactivate canonical wnt signaling and therefore plays a direct role in regulating osteogenic and adipogenic differentiation of mesenchymal progenitor cells.

- The 761 amino acid recombinant protein has a predicted molecular mass of approximately 87.9 kD. The DTT-reduced and non-reduced protein migrate at approximately 80-90 kD in SDS-PAGE.

- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at - 20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
