Stability testing for recombinant rat IL-1α. Recombinant rat IL-1α was aliquoted in 50mM Tris, pH 8.0, at 0.2 mg/ml and one aliquot was kept at 4°C (Control) and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability to induce proliferation of D10.G4.1 (mouse helper T-lymphocyte) in a dose-dependent manner. ED50 of this effect is 3-30 pg/mL.

IL-1 is a pleiotropic pro-inflammatory cytokine that exhibits biological activities supporting its role in tumor growth and metastasis. IL-1α is usually expressed as a preprotein, which is secreted only after cleavage by the calpain family of proteases. Calpains cleave pre-IL-1α and release the N-terminal propiece-IL-1α and the C-terminal secreted IL-1α. Unlike IL-1β, which is biologically active only as a secreted mature molecule, membrane-associated IL-1α, propiece-IL-1α, and mature secreted IL-1α show distinct biological activities. The propiece-IL-1α has transforming activity whereas secreted IL-1α has paracrine and autocrine activities like IL-1β. IL-1α stimulates vascularization and proliferation of cancer-associated fibroblasts thus contributes to cancer progression. IL-1α is a potent proinflammatory cytokine that mediates inflammation, microbial invasion and tissue injury. It is expressed in various immune cells, including macrophages and monocytes. It also acts as a mediator of inflammatory responses, inducing inflammation via IL-1R1-dependent pathways upon release into the tumor microenvironment. This causes persistent inflammation and stimulates the production of other proinflammatory cytokines, such as IL-6, IL-8, and TNF-α. The persistent inflammatory microenvironment leads to the deterioration of the pathological condition. IL-1α-neutralizing antibodies are currently employed in the treatment of autoimmune diseases, such as rheumatoid arthritis, as they alleviate joint inflammation via modulation of IL-1α concentrations.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
