C57BL/6 mouse splenocytes were treated with Cell Activation Cocktail (with Brefeldin A) (Cat. Nos. 423303 and 423304) for 4 hours and then fixed with Fixation Buffer (Cat. No. 420801) and permeabilized using Permeabilization Wash Buffer (Cat. No. 421002). Cells were then stained with anti-mouse CD3ε (clone 145-2C11) FITC and anti-mouse IFN-γ (clone W18272D) Spark PLUS V450™ (right) or stained with anti-mouse CD3ε (clone 145-2C11) FITC only (right).

IFN-γ is a potent multifunctional cytokine which is secreted primarily by activated NK cells and T cells. Originally characterized based on anti-viral activities, IFN-γ also exerts anti-proliferative, immunoregulatory, and proinflammatory activities. IFN-γ can upregulate MHC class I and II antigen expression by antigen-presenting cells.

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
