Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-VILIP-1 (clone A22106J). Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining with Purified anti-VILIP-1 (clone A22106J) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) and with (panel B) Purified anti-VILIP-1 (clone A22106J) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured using a 40X objective and merged (panel C and D). Scale bar: 50 µm -

Visinin-like protein 1 (VILIP-1) is a member of the neuronal calcium sensor (NCS) protein family and plays an essential role in calcium-mediated signal transduction within the human central nervous system. VILIP-1 contains EF-hand motifs that bind calcium ions, leading to conformational changes that allow it to interact with various target proteins such as adenylyl cyclases and membrane-associated signaling molecules. Through these interactions, VILIP-1 modulates cyclic nucleotide signaling and synaptic plasticity, processes fundamental to learning, memory, and neuronal development.
In Alzheimer’s disease (AD), neuronal injury and synaptic dysfunction result in the release of VILIP-1 into the cerebrospinal fluid (CSF), where it can be detected at elevated levels. CSF VILIP-1 levels correlate with biomarkers of neuronal degeneration such as total Tau and phosphorylated Tau, and can predict cognitive decline and progression from mild cognitive impairment (MCI) to AD.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
