Whole cell lysates from the indicated cell lines (15 µg total protein) were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-SOX9 antibody (clone W24113B). Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Prime-Step™ Prestained Broad Range Protein Ladder (Cat. No. 773302)

Description

SOX9 is a transcription factor of the SRY-related HMG box family that binds specific DNA motifs to control gene expression essential for chondrogenesis, testis development, neural differentiation, and maintenance of stem and progenitor cell lineages. It regulates expression by pathways such as Notch and Hedgehog and undergoes post-translational modifications, including phosphorylation and Sumoylation, that affect its nuclear localization, stability, and transcriptional potency. SOX9 is critically important for skeletal integrity and sexual differentiation; haploinsufficiency causes campomelic dysplasia and cleft palate. In adults, aberrant SOX9 expression contributes to organ fibrosis by promoting extracellular matrix production. Moreover, SOX9 dysregulation is implicated in diverse cancers, where it can enhance tumor progression, metastasis, and therapy resistance.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in Intracellular flow cytometry (ICFC), we recommend True-Phos™ Perm Buffer (Cat. No. 425401) for fixation and permeabilization.
