Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-KLF4 (clone W22026B). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining with Purified anti-KLF4 on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using Citrate Buffer (Cat. No. 420902), the tissue was incubated without (panel A) and with (panels B and C) Purified anti-KLF4 (clone W22026B) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801) (merged in panels A and B). Images were captured with a 40X objective. Scale bar: 50 µm

KLF4 (Krüppel-like factor 4) is a zinc finger transcription factor that plays a pivotal role in regulating diverse cellular processes, including proliferation, differentiation, apoptosis, and stem cell maintenance. It is one of the four Yamanaka factors (alongside Oct4, Sox2, and c-Myc) capable of reprogramming somatic cells into induced pluripotent stem cells. KLF4 can act as either a tumor suppressor or an oncogene depending on the tissue type and cellular environment. For instance, it suppresses tumor growth in colorectal and gastric cancers, while promoting tumorigenesis in breast and skin cancers. Its expression is tightly regulated at the transcriptional and post-translational levels, and it is influenced by signaling pathways such as Notch, Wnt/β-catenin, and TGF-β. Clinically, KLF4 has emerged as a potential prognostic biomarker and therapeutic target in various malignancies and its expression levels correlate with patient outcomes in cancers like renal clear cell carcinoma and glioblastoma, and it also modulates immune responses by enhancing CD8+ T cell differentiation and antitumor immunity. This dual role in tumor biology and immune regulation positions KLF4 as a key molecular bridge in cancer progression and therapy.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
