Description

KLF4 (Krüppel-like factor 4) is a zinc finger transcription factor that plays a pivotal role in regulating diverse cellular processes, including proliferation, differentiation, apoptosis, and stem cell maintenance. It is one of the four Yamanaka factors (alongside Oct4, Sox2, and c-Myc) capable of reprogramming somatic cells into induced pluripotent stem cells. KLF4 can act as either a tumor suppressor or an oncogene depending on the tissue type and cellular environment. For instance, it suppresses tumor growth in colorectal and gastric cancers, while promoting tumorigenesis in breast and skin cancers. Its expression is tightly regulated at the transcriptional and post-translational levels, and it is influenced by signaling pathways such as Notch, Wnt/β-catenin, and TGF-β. Clinically, KLF4 has emerged as a potential prognostic biomarker and therapeutic target in various malignancies and its expression levels correlate with patient outcomes in cancers like renal clear cell carcinoma and glioblastoma, and it also modulates immune responses by enhancing CD8+ T cell differentiation and antitumor immunity. This dual role in tumor biology and immune regulation positions KLF4 as a key molecular bridge in cancer progression and therapy.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).

Antigen Details

Structure
The human KLF4 protein is 513 amino acids length and has a predicted molecular weight of approximately 55 kDa.
Distribution

KLF4 is mainly located in the nucleus but may also be localized in the cytoplasm.

Interaction
• p300/CBP (CREB-binding protein): Acts as a transcriptional co-activator; KLF4 interacts with these to enhance gene expression
• Prmt1 (Protein arginine methyltransferase 1): Methylates KLF4, influencing its role in cell fate decisions and preventing primitive endoderm differentiation
• OCT4 (POU5F1) and SOX2: Core pluripotency factors; KLF4 interacts with them during somatic cell reprogramming
• MUC1: Interacts via KLF4’s C-terminal domain, potentially influencing epithelial cell signaling
• MEIS2 and PBX1: Homeobox transcription factors involved in development; KLF4 binds to specific isoforms of these proteins
