Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-SDHA (clone W23126E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat.     IHC staining with Purified anti-SDHA (clone W23126E) was performed on FFPE human heart tissue following antigen retrieval using 1X Citrate buffer (pH 6.0) (Cat. No. 420902). The tissue sections were incubated with 5 µg/mL of Purified anti-SDHA (clone W23126E) (panels B and C) followed by Alexa Fluor® 594 Goat anti-rat (Cat. No. 405422) (red) or secondary antibody only (panel A). Nuclei were counterstained with DAPI (Cat. No. 422801) (blue) (merged in panels A and C). The image was captured with a 40X objective.     HeLa cells treated with mock shRNA (panel B) or knockdown with transiently transfected SDHA shRNA (panel C) were fixed with fixation buffer (Cat. No. 420801) for 10 minutes. Then, permeabilized with ice-cold methanol and blocked with 5% FBS for 1 hour at room temperature. Cells were then intracellularly stained with isotype control (Cat. No. 400602) (panel A) or stained with Purified anti-SDHA (clone W23126E), followed by incubation with Alexa Fluor 594 Goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Then, cells were co-stained with Alexa Fluor® 488 anti-GFP Antibody (Cat. No. 338008) to detect expression of shRNA reporter for 1 hour at room temperature (panel C). Description

Succinate dehydrogenase (SDH), commonly known as respiratory complex II, is an important enzyme of mitochondria in the citric acid cycle and electron transport chain, and consists of four subunits SDHA, SDHB, SDHC, and SDHD. SDHA, like other SDHx genes, can function as a tumor suppressor gene. Recently, studies have indicated that mutations in SDHA contribute to various disorders such as hereditary pheochromocytoma-paraganglioma (PPGL) syndrome, mitochondrial complex II deficiency, gastrointestinal stromal tumors, Leigh syndrome, dilated cardiomyopathy, and neuroendocrine tumors.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

This antibody has been tested in knockout/knockdown systems for WB and ICC.

This antibody (clone W23126E) does not cross-react with mouse SDHA on Western Blot (WB).

For immunocytochemistry (ICC), we recommend fixation with 4% PFA fixation followed by permeabilization with 0.5% Triton X-100 or ice cold-methanol, or ice-cold methanol fixation.

For immunohistochemistry on FFPE tissues (IHC-P), antigen retrieval with Citrate Buffer (Cat. No. 420901) or Tris-EDTA pH 9.0 (Cat. No. 422704) is recommended.
