Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel. A549 cells were untreated or treated with human TGF-β1 (10 ng/mL) (Cat. No. 781804) and human IL-1β (5 ng/mL) (Cat. No. 579404) for 48 hours. Then, the gel was transferred to a PVDF membrane, and probed with the Purified anti-HMGA2 (clone W23012E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat.     A549 cells were untreated (mock) and treated with human TGF-β1 (10 ng/mL) (Cat. No. 781804) and human IL-1β (5 ng/mL) (Cat. No. 579404) for 48 hours. The mock and treated A549 cells were fixed with Fixation buffer (Cat. No. 420801) for 10 minutes, permeabilized with TritonX-100 then blocked with 5% FBS for 1 hour at room temperature. Cells were then intracellularly stained with Purified Rat IgG2b, κ Isotype Control Antibody (Cat. No. 400602) (panels A, D) or stained with Purified anti- HMGA2 (clone W23012E) (panels B, E), followed with Alexa Fluor® 488 Goat anti-rat IgG (Cat. No. 405418) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801) and actin cytoskeleton were labeled by Flash Phalloidin™ Red 594 (Cat. No. 424203). The image was captured with a 40X objective. Description

HMGA2, also known as the high-mobility group AT-hook 2, is a nonhistone transcriptional regulator belonging to the high mobility group (HMG) protein family with AT-hook DNA binding motif. HMGA2 is highly expressed during early mammalian embryogenesis but is hardly detected in normal adult tissues. Recently, it was discovered that HMGA2 is upregulated in a variety of tumors including gastric cancer, ovarian cancer, non-small cell lung cancer and colorectal cancer, suggesting that it has a role in controlling tumor progression. The cancers with overexpression HMGA2 have a significantly stronger response to double strand breaks in DNA caused by radiation therapy and certain chemotherapy treatments. HMGA2 also has been implicated in diet-induced obesity in the gene knockout study of the mouse.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

The clone cross-reacts with mouse and rat in western blot (WB). 

For immunocytochemistry (ICC), we recommend 4% PFA fixation (Cat. No. 420801) followed by permeabilization with Triton X-100 or ice-cold methanol.

For immunohistochemistry on FFPE tissues (IHC-P), antigen retrieval with Citrate Buffer (Cat. No. 420901) or Tris-EDTA pH 9.0 (Cat. No. 422704) are recommended.
