C57BL/6 mouse splenocytes were stimulated with (left) or without (right) recombinant human TGF-β1 and recombinant mouse IL-6 for 3 days. Cells were surface stained with anti-mouse CD4 APC, fixed and permeabilized, then intracellularly stained with anti-mouse AhR PE (clone W16012C).

The aryl hydrocarbon receptor (AhR) is a basic helix-loop-helix transcription factor that is activated by multiple ligands such as TCDD. It is expressed in many immune cells and performs an important role in immune regulation. AhR is induced under Th17-polarizing conditions. In its inactive state, AhR is present in the cytoplasm, but upon ligand binding, it translocates to the nucleus. Activation of AhR is tightly controlled by ubiquitination, ligand metabolism, and AhR complex disruption.

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
