LEW/Ss rat peritoneal macrophages were surface stained with anti-rat CD11b/c (clone OX-42) FITC. Cells were then fixed and permeabilized with Cyto-Fast™ Fix/Perm Buffer (Cat. No. 426803) and intracellularly stained with anti-rat CD68 recombinant (clone QA20A71) Brilliant Violet 605™ (left), or mouse IgG1, κ Brilliant Violet 605™ isotype control (right).

CD68, also known as macrosialin, is a 110 kD integral membrane glycoprotein from the lysosomal-associated membrane protein (LAMP) family. It is predominantly expressed on the intracellular lysosomes of monocytes and macrophages, and is considered a pan-macrophage marker in the rat. CD68 may have functions relating to phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions.

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Brilliant Violet 605™ excites at 405 nm and emits at 603 nm. The bandpass filter 610/20 nm is recommended for detection, although filter optimization may be required depending on other fluorophores used. Be sure to verify that your cytometer configuration and software setup are appropriate for detecting this channel. Refer to your instrument manual or manufacturer for support. Brilliant Violet 605™ is a trademark of Sirigen Group Ltd.

Clone QA20A71 can completely block the binding of parental clone to target cells. Both Cyto-Fast™ Fix/Perm Buffer Set and True-Nuclear™ Transcription Factor Buffer Set work well. This clone does not react to human and mouse.
