Biotinylated recombinant human SIRPα-Fc chimera (Cat. No. 561014; black circles) binds to immobilized recombinant human CD47. Ultra-LEAF™ Purified anti-human CD47 (clone W24220J, purple squares) inhibits the binding in a dose-response manner, whereas Ultra-LEAF™ Purified rat IgG2a, κ Isotype Control (Cat. No. 400543, gray triangles) has no inhibitory effect. This antibody blocks the binding of 500 ng/mL biotinylated recombinant human SIRPα-Fc Chimera to 1.0 µg/mL of immobilized recombinant human CD47. The ND50 range to observe this effect is: 28.0 – 285.0 ng/mL. Main competitor ND50 = 351.1 ng/mL.

CD47, also known as Rh-associated protein, gp42, integrin-associated protein (IAP) and neurophilin, is a 42-52 kD member of the immunoglobulin superfamily containing a five-pass transmembrane attachment. Two splice variants have been described for the cytoplasmic tail: a shorter form, expressed in bone-marrow-derived cells, endothelial cells, and fibroblasts; and a longer form, expressed mainly in neural tissues. CD47 expression is widely distributed in hematopoietic cells, including thymocytes, T cells, B cells, monocytes, platelets, and erythrocytes as well as epithelial cells, endothelial cells, fibroblasts, and neural tissues. CD47 functions as an adhesion molecule and a thrombospondin receptor and is non-covalently associated with β3 integrins CD51/CD61, CD41/CD61. Thrombospondin is a ligand for CD47; in the absence of CD47, mice present defects in host defense and β3 integrin-dependent ligand binding, migration, and cellular activation. CD47 is also part of the Rh complex on erythrocytes.

Each lot of this antibody is quality-controlled tested by blocking the binding of 1 μg/mL immobilized recombinant human CD47 (Cat. No. 785406) to its ligand biotinylated recombinant human SIRPα-Fc Chimera (Cat. No. 561016) at 500 ng /mL. ND50 range: 28.0 – 285.0 ng/mL. It is recommended that the reagent be titrated for optimal performance for each application.
