Description

Ly-6A/E, also known as Sca-1, is an 18 kD member of the Ly-6 multigene family. Ly6A/E is a glycosylphosphatidylinositol (GPI)-linked protein expressed on hematopoietic stem cells. In mice expressing the Ly-6.2 haplotype (e.g., AKR, C57BL, C57BR, DBA/2, SJL, SWR, and 129), Ly-6A/E is also expressed on peripheral B lymphocytes and thymic and peripheral T lymphocytes. Strains expressing the Ly-6.1 haplotype (e.g., BALB/c, CBA, C3H/He, DBA/1, and NZB) have low Ly-6A/E expression on resting peripheral lymphocytes. The expression of Ly-6A/E on lymphocytes is upregulated upon activation from both Ly6.1 and Ly6.2 haplotype mice. Ly-6A/E is thought to be involved in the regulation of both T and B cell responses.

Recommended Usage

Flexi-Fluors™ are provided at a standard 0.2 mg/mL concentration. We recommend titrating this reagent to determine the optimal concentration for each application. For many flow cytometry applications, conjugated antibodies perform well at concentrations ranging from 0.03 to 1.0 µg per million cells in 100 µL. We recommend testing a range of concentrations starting from 10 µg/mL.

For example, make five 1:1 serial dilutions of the 0.2 mg/mL antibody. Add 5 µL of each dilution (including the undiluted antibody) to 100 µL of cells (at 107 cells/mL) to test six concentrations -- 1.0, 0.5, 0.25, 0.125, 0.06, and 0.03 µg per million cells in 100 µL volume. Compare staining patterns or create a titration curve using the MFI or staining index to determine the optimal concentration.

* Spark YG™ 581 has a maximum excitation of 562 nm and a maximum emission of 581 nm.

Application Notes

The D7 antibody has been reported to induce T cell activation and inhibit TCR-induced IL-2 production. Additional reported applications (for the relevant formats) include: Western blotting1,2, immunoprecipitation1, in vitro lymphocyte activation3-6, induction of redirected lysis7, induction of T cell inhibitory signalling8, immunofluorescence9, and immunohistochemical staining of acetone-fixed frozen sections13 and Bouin-fixed, paraffin-embedded samples9.

The two Sca-1 recognizing clones D7 and E13-161.7 have been shown to bind distinct epitopes due to the inability of D7 to block the binding of E13-161.7.14

How do I determine the optimal concentration for using my Flexi-Fluor? How should I titrate my antibody?

Yes, you can order multiple vials of the same Flexi-Fluor products.  We cannot guarantee, however, that these vials will be bottled from the same lot. For bulk single-lot orders, contact our Custom Solutions team.

What is the expiration date of my Flexi-Fluor?

What if my requested antibody-dye combination doesn't perform as expected?

Flexi-Fluor™ products have been carefully selected by our scientists for their excellent conjugation properties and broad compatibility. In the rare case that an antibody-dye combination does not perform as expected due to the unique characteristics of a specific pairing, please contact our Technical Service team. We will work with you to identify alternative options to meet your panel requirements.
