Human epidermoid carcinoma cell line, A431, was stimulated with 50 ng/mL of recombinant human IFN-γ (Cat. No. 713906) for 48 hours (panels A and C) or left unstimulated (panel B). Cells were fixed, permeabilized, and then stained with 5 µg/mL of purified anti-human IDO1 (clone W20099F) (panels A and B) or purified rat IgG2b, κ isotype control (panel C), followed by anti-rat IgG Alexa Fluor® 594. Nuclei were counterstained with Hoechst 33342. Cells were imaged using a 40X objective. Scale bar = 20 µm

IDO1, also known as indolamine 2,3-dioxygenase, indole 2,3-dioxygenase, and indoleamine-pyrrole 2,3-dioxygenase, is a ubiquitously expressed cytoplasmic protein with a predicted molecular weight of approximately 45 kD. It is one of the best-known IFN-γ inducible genes. The IDO1 enzyme catalyzes the degradation of the essential amino acid L-tryptophan to N-formylkynurenine. IDO1 plays an important role in host defense against both intracellular and extracellular pathogens and contributes to the special immunosuppressive status of immune-privileged sites such as the brain, eyes, kidneys, and placenta.
