Whole cell extracts (15 µg total protein per lane) from the indicated cell lines or recombinant protein (Recombinant Human FN-1, Cat. No. 775306) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Fibronectin (clone C6F10) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
HeLa cells (negative cell line, panel A) and HepG2 cells (positive cell line, panel B) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-Fibronectin (clone C6F10), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm -
HepG2 cells (positive cell line, filled histogram) and HeLa cells (negative cell line, open histogram) were fixed and permeabilized using True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) and intracellularly stained with Purified anti-Fibronectin (clone C6F10) or Purified Mouse IgG1, κ isotype control (dashed histogram) (Cat No. 400102) followed by Alexa Fluor® 647 anti-mouse IgG1 Antibody (Cat. No. 406618).

Fibronectin is a glycoprotein that plays a critical role in cell adhesion, growth, migration, and differentiation. It is a major component of the extracellular matrix (ECM), where it helps to organize other matrix proteins and anchor cells via integrin receptors. Fibronectin is essential during embryonic development and wound healing, facilitating tissue morphogenesis and repair by guiding fibroblast and endothelial cell movement. It also contributes to mechanotransduction and regulates cellular signaling pathways involved in survival and proliferation. Dysregulation of fibronectin expression or structure has been implicated in fibrosis, cancer metastasis, and chronic inflammation.

ICC, ICFC - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
