Tyrosine-protein kinase receptor AXL (also known as Ark, Tyro7, and UFO), TYRO-3 (also known as Brt, Dtk, Rse, Sky, and Tif), and MERTK (also known as Eyk, Nym, and Tyro12) are included in the small family of TAM receptor tyrosine kinase. The TAM family is characterized by a combination of two immunoglobulin-like (Ig) domains and dual fibronectin type III (FNIII) repeat domains in the extracellular region, a transmembrane domain and a cytoplasmic tyrosine kinase domain.
One of the ligands of the TAM family, growth arrest specific gene 6 (Gas6), are secreted proteins and mediate their action through binding to and activating of AXL. AXL can also be activated by ligand-dependent dimerization, ligand-independent dimerization, and interaction between two monomers on neighboring cells and heteromeric dimerization with a non-TAM receptor.
AXL regulates multiple biological processes in cancer. Activated AXL, by binding to Gas6, promotes various downstream signaling pathways such as PI13k/Akt, JAK, and RAS/MEK/ERK pathways that can affect cancer cell proliferation, survival, migration, invasion, angiogenesis, stem cell maintenance, and immune suppression.
