When immobilize human BMP-3b/GDP-10 binds to recombinant human BMPR1B/ALK-6 in a dose-dependent manner. The ED50 for this effect is 0.5 – 3.5 µg/mL. -
Stability Testing for recombinant human BMPR1B/ALK-6: Recombinant human BMP-3b/GDP-10 was aliquoted in 0.1% TFA, 10% acetonitrile at 0.2 mg/mL. One aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). The samples were evaluated in a binding assay with immobilized human BMP-3b/GDP-10 and different concentration of biotinylated recombinant human BMPR1B/ALK-6 that binds in a dose-dependent manner. The ED50for this effect is 0.5 – 3.5 µg/mL. -
Western blot analysis: COV434 cells starved overnight and then treated for 1 hour with the following conditions: no treatment (lane 1); 1 ng/ml recombinant human BMP-9 (lane 2); various concentrations of recombinant human BMP-3b/GDP-10 (lane 3: 0.2 µg/ml, lane 4: 0.1 µg/ml, lane 5: 0.05 µg/ml); and 0.2 µg/ml recombinant human BMP-3b/GDP-10 from a competitor (lane 6). Lysates were subjected to SDS page followed by western blot with an anti-SMAD1 Phospho (S463/S465)/SMAD9 (S465/S467) (clone A17019A) (Cat. No. 610402) or purified anti-β-actin (clone Poly6221) (Cat. No. 622101).

Bone Morphogenetic Protein 3b (BMP-3b), also known as Growth Differentiation Factor 10 (GDF-10), is a member of the Transforming Growth Factor-beta (TGF-β) superfamily. Although both BMP-3 and BMP-3b belong to the BMP family, they exhibit distinct structural characteristics. BMP-3b shares approximately 82% amino acid sequence identity with BMP-3 in the mature region and 37% in the pro-domain. Like other BMP family members, BMP-3 is synthesized as an inactive propeptide precursor, which dimerizes and is subsequently processed into its mature form. Functionally, BMP-3 acts as an antagonist to BMP-2-induced osteogenic signaling, inhibiting the differentiation of osteoprogenitor cells into osteoblasts. BMP-3 antagonizes the BMP type II receptor ActRIIB and serves as a negative regulator of bone density. BMP-3 expression is observed in osteoblasts at erosion sites during the late stages of inflammatory arthritis and has been associated with colorectal cancer and gastric carcinoma. Overexpression of BMP-3 during skeletal development modifies endochondral bone formation resulting in spontaneous rib fractures. BMP-3b is expressed in various tissues, including the brain, skeletal tissue, and adipose tissue. It plays key roles in neurodevelopment, bone formation, adipogenesis, metabolic regulation, tissue homeostasis, and oncogenesis. BMP-3b treatment has been shown to promote neuronal growth and improve functional recovery in a mouse model of stroke. Additionally, in a neonatal mouse model of hypoxia-ischemia-induced encephalopathy, BMP-3b administration enhanced survival outcomes. BMP-3b also acts as a negative regulator of osteoblast differentiation.

- The 112 amino acid recombinant protein has a predicted molecular mass of approximately 12.7 kD. The DTT-reduced protein and the non-reduced protein migrate at approximately 10 - 15 and 20 - 30 kD by SDS-PAGE, respectively. The N-terminal amino acid is Met.

Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
