The Brightness Index is a relative indication of fluorescence intensity above the background for each fluorophore antibody conjugation (1=dim, 5=brightest). These values should be used as a general reference tool, as several factors can influence a fluorophore's brightness, such as instrument settings, the density of the antigen target, and testing conditions.
The fluorophores listed below are ordered by:
- Brightness - The fluorophores are grouped by brightnesses levels, with rank 5 fluorophores being displayed first.
- Excitation laser wavelength - Within a brightness ranking, fluorophores with the lowest excitation laser wavelength will be listed first. For example, StarBright™ UltraViolet 740 and Spark PLUS B488™ are both brightness level 5, but the StarBright™ UltraViolet 740 listing comes first as its excitation laser is the UV 355 nm laser, whereas Spark PLUS B488™ utilizes the Blue 488 nm laser. PE and its tandems will utilize the yellow/green laser as their main excitation laser.
- Emission wavelength - If fluorophores are the same brightness level and share the same excitation laser, then the fluorophore with the lower emission wavelength will be listed first. For example, PE (Emmax 575 nm) appears before PE/Dazzle™594 (Emmax 610 nm).
